三、設備可(kě)以實現功能/Functions of SPR
　　儀器 SPR 是基于表面等離子體共振（SPR）技術來(lái)實時(shí)跟蹤生物(wù)分(fēn)子間的(de)相互作用(yòng)，不需任何标記物(wù)的(de)技術。該設備通(tōng)過固定波長(cháng)，測量角度反射光(guāng)譜獲得(de)分(fēn)子相互作用(yòng)信息；也(yě)可(kě)固定入射角度，測量分(fēn)子相互作用(yòng)動力學曲線。SPR可(kě)進行：1、動力學常數的(de)測定；2、樣品濃度的(de)測定；3、分(fēn)析相互作用(yòng)模式；4、複合物(wù)功能分(fēn)析。

The instrument SPR is based on Surface Plasmon Resonance (SPR) technology to monitor the interaction of biomolecules in real time. The molecular interactions information is obtained by measuring the angular reflection spectroscopy at a fixed wavelength; or measuring the kinetic of molecular interactions through reflectivity intensity changes at a fixed incident angle. SPR is able to perform the determination of kinetic constant, detection of sample concentration; analysis of the interaction modes and functional analysis of complex.

五、基本原理(lǐ)/Principle
　　如圖所示，SPR 分(fēn)子相互作用(yòng)儀器的(de)原理(lǐ)是波長(cháng) 632.8 nm 的(de)激光(guāng)形成的(de) p 偏振光(guāng)，到達棱鏡後激發金屬表面的(de)表面等離子體發生共振。通(tōng)過改變入射光(guāng)角度 θ，實時(shí)檢測反射光(guāng)強度的(de)變化(huà) R，根據反射光(guāng)強度變化(huà) ΔR 或共振角度的(de)變化(huà) Δθ，實現分(fēn)子相互作用(yòng)的(de)檢測；也(yě)可(kě)以通(tōng)過固定角度，測量反射光(guāng)強的(de)變化(huà)曲線獲得(de)分(fēn)子相互作用(yòng)動力學常數。

As shown in the figure, the laser at the wavelength of 632.8 nm passes through a polarizer to become a transverse magnetically polarized beam and is coupled to glass prism to excite surface plasmon resonance of sensor chip. The reflected light intensity R is detected in real time by the changes of incident light angle θ. The detection of molecular interactions can be achieved by the changes of ΔR or Δθ; the molecular interaction kinetic constants can also be obtained by measuring the reflectivity changes at a fixed angle.
