3 Functions of SPR
The instrument SPR is based on Surface Plasmon Resonance (SPR) technology to monitor the interaction of biomolecules in real time. The molecular interactions information is obtained by measuring the angular reflection spectroscopy at a fixed wavelength; or measuring the kinetic of molecular interactions through reflectivity intensity changes at a fixed incident angle. SPR is able to perform the determination of kinetic constant, detection of sample concentration, analysis of the interaction modes and functional analysis of complex.

5 Principle

As shown in the figure, the laser at the wavelength of 632.8 nm passes through a polarizer to become a transverse magnetically polarized beam and is coupled to glass prism to excite surface plasmon resonance of sensor chip. The reflected light intensity R is detected in real time by the changes of incident light angle θ. The detection of molecular interactions can be achieved by the changes of ΔR or Δθ; the molecular interaction kinetic constants can also be obtained by measuring the reflectivity changes at a fixed angle.
